To date, the mechanisms of action of plasma treatment on biological tissue are only incompletely understood. The aim of this project is to elucidate the mechanisms of action of dielectrically impeded atmospheric pressure plasma on the outer skin barrier and in particular on the lipids of the stratum corneum. Although the lipids of the stratum corneum are of great importance for the barrier function of the skin and some important findings have recently been published, particularly with regard to mutations in the filaggrin gene and the associated diseases such as ichthyosis and atopic dermatitis, a great deal of basic research is still required to fully understand the relationships.

The aim of this project is to contribute to the development of physically based strategies for the use of plasma devices and their parameter limits as well as to the systematic investigation of skin barrier function. Of particular medical interest is the comparison of the lipid composition of humans and mice with filaggrin mutations and the influence of plasma on lipids in a comparison of human and mouse samples. In order to do justice to the complexity and interdisciplinary nature of the topic, one partner each from the medical, physical and technical fields are participating in the application.

The following spectroscopic diagnostic methods will be used: X-ray photoelectron spectroscopy (XPS), ultraviolet photoelectron spectroscopy (UPS) and the particularly surface-sensitive "Metastable Induced Electron Spectroscopy" (MIES).
